Zusammenfassung

Grapevine (Vitis vinifera L.) is one of the most economically important crops worldwide. However, it is susceptible to several diseases caused by a wide range of organisms. Among them white root rot caused by Armillaria mellea (Vahl:Fr.) P. Kumm. and trunk diseases are particularly serious because there is no effective control method against. A innovative approach is to generate a transgenic grapevine with modificated content and/or composition of its lignins to increase tolerance. Therefore, the aim of this dissertation is to assess the potential of this strategy through the characterization of a grapevine peroxidase gene (VviPrx73) and its expression in transgenic tobacco (Nicotiana tabacum L.) plants to analyze their phenotypical changes. Our results suggest an involvement of VviPrx73 in cell wall lignification, nevertheless its expression in transgenic tobacco plants has not been found to be related to a phenotype of interest in terms of defense against pathogens. Apart from the significant increase in apoplastic peroxidase activity against coniferyl and sinapyl alcohols observed in all transgenic lines, one of the transgenic lines has shown phenotypic differences compared to controls and these differences do not correlate with the presence of VviPrx73.
